Tempero Bio, a clinical-stage biopharmaceutical company, is revolutionizing addiction treatment by securing $70 million in Series B funding to advance clinical trials for substance use disorder, alcohol use disorder, and cocaine use disorder.
The investment, led by 8VC with contributions from Aditum Bio, Khosla Ventures, and other investors, will propel the company’s breakthrough drug, TMP-301, through advanced clinical trials.
Designed to target the brain’s addiction pathways, TMP-301 is a next-generation therapy aimed at addressing the root causes of substance use disorder. As a metabotropic glutamate receptor 5 (mGluR5) negative allosteric modulator (NAM), the drug has shown significant promise in preclinical studies for alcohol use disorder, cocaine use disorder, and other addictions.
In early trials, over 80 healthy participants demonstrated a favorable safety profile, with PET imaging confirming its strong receptor targeting.
“TMP-301 is a novel therapy that helps prevent relapse by targeting the underlying biology of addiction. This financing will allow us to advance TMP-301 through key proof of concept phase 2 human studies,” said Ricardo Dolmetsch, President and Chief Scientific Officer of Tempero Bio.

Following this, a second Phase 2 trial will specifically target cocaine use disorder, a condition for which no FDA-approved addiction treatment currently exists. The National Institute on Drug Abuse has backed this research, highlighting its potential to address an urgent public health crisis.
“Substance use disorders affect 48 million Americans and contribute to more than 100,000 deaths per year. We urgently need more effective treatments to help patients and families with these diseases,” said Ricardo Dolmetsch.
Addressing a Growing Addiction Epidemic
The latest survey data paints a troubling picture. In 2023, 28 million Americans suffered from alcohol use disorder, while 29 million struggled with substance use disorder. Among them, 1.3 million battled cocaine use disorder without an approved medical solution.
Yet, nearly 13 million people sought addiction treatment, underscoring the pressing need for better therapeutic options. Tempero Bio aims to bridge this treatment gap by advancing science-driven solutions through clinical trials.
Strengthening its leadership, Tempero Bio has appointed Dr. John Wagner as Chief Medical Officer. Over two decades of experience in addiction treatment research and clinical trials, he has spearheaded over 150 first-in-human trials and contributed to several blockbuster drug launches.
His previous roles at Koneksa Health, Cygnal Therapeutics, Takeda, and Merck, along with his involvement in the National Institutes of Health Biomarkers Consortium, make him a key asset in guiding TMP-301 through regulatory approval.
With this new funding, the company is positioned to redefine addiction treatment and offer hope to millions struggling with alcohol use disorder and cocaine use disorder.
